... so what's the most effective place and time to buy a copy, in terms of encouraging booksellers to order a stack of books from your publisher to display prominently?

Pre-order now or just before release or just after? Local independent, online store, brick and mortar chain?

My local indie bookstore says that preorders placed more that 3 months pre-release get kicked out of the system, so I'm there June first. Of course, I also ordered a copy online, so I'm covered for the kitten-compulsion.

Honestly, your individual order won't change the initial orders from B&N or Borders, both of which will have their order size determined by corporate (here's hoping it's awesome). [livejournal.com profile] catnip13 is right about waiting for June if not ordering through one of the links above; I'd recommend your local independent if you have one, because they always need the sales.
